Order of the Stick - funny, plenty of plot, loveable characters
As a result, Erfworld, I guess - more 'plot'-based, nice art
I read Chugworth Academy because of Jello's sig - funny, but sometimes too much fanservice, slow-moving plot

Order of the Stick: my personal favourite. Great plot, excellent characters, and still manages to deliver a humorous punchline every episode (there are exceptions, but they are so great story-wise that a slapstick would just ruin them). Also, great and (mostly) mature community.

Erfworld: shares the site with OotS. It's just starting, but looks nice. Pun-a-riffic extravaganza, and an interesting plot.

The Noob: slow updates, but a funny commentary on the world of MMORPGs.

8-Bit Theater: does it really need an explanation? Let's just add that contrary to what people say, I believe that it actually gets BETTER in time, and that nuklearpower.com's forum is awful, don't read it.

Commissioned: No real plot, a webcomic made by a professional artist, describing either events that him and his friends (or their RPG characters) face, or just random stuff. Updates daily.

VG Cats: maybe low-brow, but 100% hilarious.

I discovered this one a few days ago. It's fairly new but it's funny.
